The Evaluation on the Efficacy of STR immunity materials

STR biotech has various in-vivo animal experimental study cases in order to evaluate the efficacy and effect of the immunity materials and is performing the research and monitoring evaluating the efficacy and effect fit for the characteristics of the materials.

1) The Life-Expanding Effects in the case of Animal transplanted with Cancer Cells

Followings are experiment results proving its virtue against cancer by the life expanding effects of STR immunity materials by putting Beneauri-AA into the abdominal cavity of ICR mice transplanted with Sarcoma 180 cancer cell strains.

Experiment has been conducted to confirm the life-expanding effects of STR immunity materials by dividing mice into 3 groups, a normal group, a group of mice led to cause multiple cancers (control group) by transplanting Sarcoma-180 cell into the abdominal cavity of mice and a group of mice with transplanted cancer cells injected with Beneauri-AA into their abdominal cavity at 10mg/kg body weight/day.

The life-expanding effects were evaluated by observing the mortality during the period of for 30 days after injecting Beneauri once per day for 14 days into the abdominal cavity of mice transplanted with cancer cells.

The excellent life-expanding effects could be proven by the experiment results that all the mice in the control group led to cause multiple cancers died within the trial period with an average period of survival for 10.7 days but that only one out of 10 mice in the group injected with Beneauri-AA died within the trial period with an average period of survival for 28.5 days.

2) Improvement Effects in the mortality from the bacteria infection in the experiment of animals infected with Salmonella Typhimurium
Followings are results from the observation of the improvement of the mortality in the experiment of Balb/c mice infected with Salmonella typhimurium after oral administration of Benephelli-BRB for 14 days.

Mechanism of action should be first confirmed in the immunity materials

STR biotech has various in-vitro cell experimental cases in order to evaluate the mechanism of action for the immunity materials and is performing the evaluation and monitoring of the mechanism of action fit for the characteristics of the materials.

1) Activation of Macrophages
Followings are experiment results if Benephelli-BRB having an immunity boosting and adjustment efficacy can activate macrophages that are the innate immune system cells.

We found from the activation efficacy of macrophage in the abdominal cavity of C57BL/6 mouse that the macrophages in the abdominal cavity were being activated to the almost same level that done by addition of LPS at the very low concentrations of 1ug/mL.

The macrophages are innate immune system cells devoting to eating viruses or pathogens directly. As a result, the immunity will be boosted when the macrophages become activated.
Benephelli-BRB having an immunity boosting and adjustment efficacy is exceptionally effective in boosting the innate immunity responding instantly and fast to the bacteria and viruses coming into our body.

2) Activation of Dendritic Cells
Followings are experiment results showing that Beneauri-AA having an immunity boosting and adjustment efficacy activates dendritic cells by contributing to the maturation of dendritic cells that are carrying out the role to bridge an innate immune system and an acquired immune system.

  • It was found that the dendritic cells were growing to matured ones by the treatment of Beneauri-AA at the very low concentrations of 1mg/mL
  • according to the result analyzing the phenotype of the matured dendritic cells by FACS induced by Beneauri-AA matured by treating LPS induced matured dendritic cells grown to matured dendritic celles (mDC) by treating LPS and beneauri-AA
  • in the inmmatured dendritic cells (imDC) differentiated by treating GM-CSF and IL-4 in the monocyte separated from human peripheral blood.

Beneauri-AA has been found to be exceptionally effective not only in activating the dendritic cells through the differentiation and maturation to the matured dendritic cells, but also in boosting the acquired immunity by contributing to the activation of the acquired immunity through affecting to the activation and proliferation of T cells through activated dendritic cells.

The background that the evaluation on safety should be made

It is true that there are concerns about the fact that serious inflammatory reaction is developed when the immunity reaction is increased.

LPS (lipopolysaccharide) is a typical immunity activation substance activating macrophages that are the innate immune system cells and dendritic cells by involving in the maturation process of dendritic cells and also directly activating and proliferating B cells that are the acquired immune system cells non-specifically. But LPS is, as a substance causing sepsis by the infection of pathogenic microorganism, makes an animal lead to death by injecting only a small portion of it causing serious damages to organs such as liver, kidney, lung, etc. together with a systemic inflammation.

Therefore, the evaluation on the safety should be certainly made in the process of the development of immunity boosting (Adjustment) materials.

1. The Inhibition of Systemic Inflammatory Reaction and Damage to Organs in the case of Animal with Sepsis caused by LPS
Followings are the results showing that Benephelli-BRB is effective in lowering the concentrations of TNF- in the accelerated blood and inhibiting the damage to organs such as liver, etc. in the experiment of Balb/c mice with sepsis caused by LPS 5mg/kg, Galactosamine 700mg/kg.

Significantly improved results could be observed in the group injected with Benephelli compared to the group with Sepsis caused by LPS in the analysis of TNF-α that is an indicator related to systemic inflammation.

It was observed that the GOT/GPT concentrations in blood increased in the group with sepsis caused by LPS has been lowered in the group injected with Benephelli when compared with the indicators related to liver damage.

As a result of injecting and orally administering Benephelli-BRB into the abdominal cavity, it could be verified that TNF-alpha concentrations in the accelerated blood from sepsis caused by LPS/Galactosamine have been lowered and that the damage to organs such as liver, lung, kidney, etc. has also been inhibited.

Benephelli-BRB like LPS does activates macrophages and dendritic cells.
But the points differentiating Benephelli-BRB from LPS with a severe toxicity are that it can't activate and proliferate B cells directly and also that it inhibits the systemic inflammatory reaction and damage to organs such as liver, kidney, lung, etc. caused by LPS while LPS causes sepsis.
