Anti-oxidant effect

DPPH radical scavenging assay
Free radical is oxygen in unstable state unlike oxygen that we inhale for breathing and is also called
harmful oxygen, which is oxygen to be produced excessively by environmental pollution and chemicals,
UV rays, blood circulation disorder, stresses, etc. Such excessively produced oxygen causes oxidation
in our body, so it makes various problems and causes skin aging as well.

SOD assay : Anti-oxidation test using Superoxide dismutase(SOD)
SOD is an enzyme involved in elimination of O2-(superoxide) in a living body and materials bearing SOD-like
activities are explored to inhibit free radical because free radical generated in a living body causes oxidative
disorders in the body.

Anti-inflammatory effect
NO assay : Checking generation of nitric oxide(NO) generated by inflammatory response.
NO is one of free radicals which plays an important role to cause inflammation. Therefore, the inhibitory
effect on NO generation can be said as same as anti-inflammatory effect.

Western blot assay

• Checking expression of iNOS generated by inflammatory response
• Checking expression of COX-2 generated by inflammatory response

iNOS (inducible nitric oxide synthase) expressed by LPS stimulation generates plenty of NO and
the cytotoxicity caused by this deepens the inflammatory response.
An enzyme called CPX-2 is generated by NO, which produces inflammatory agent called PGE₂
(Prostaglandin E₂).

Whitening effect

Tyrosinase is a multifunctional, copper-containing oxidase that catalyzed three distinct reactions of melanin synthesis, the hydroxylation of tyrosine to 3,4-dihydroxyphenylalanine(DOPA), the oxidation of DOPA to dopaquinone, and the conversion of 5,6-dihydroxyindole to melanochrome.
The enzymatic oxidation of L-tyrosine to melanin is of considerable importance because melanin has many functions. Recently, tyrosinase inhibitors have become important in cosmetic and medical products in relation
to hyperpigmentation.

Anti-bacterial effect
As modern civilization is developed, various skin diseases such as atopic dermatitis, allergy, etc. are increased and the importance of skin infectious bacteria for such skin diseases also gets higher. Anti-bacterial activity against such skin infectious bacteria stands out as an important issue for maintenance of healthy and beautiful skin.

MIC test
It is one of major test methods to evaluate antibacterial activity of a testing material by minimum inhibitory concentration method. It is a testing method to check the minimum concentration of a testing material inhibiting growth of testing strains by measuring turbidity after engrafting and culturing the testing strains in culture media treated with the testing materials by concentration.

Paper disk diffusion test
It is also called agar diffusion method and can measure antimicrobial activity in a short period. When the growth of bacteria is inhibited in a circle around paper disk, microorganism clear zone is created and it is a testing method simply to compare the size of such a circle.

Challenge test

It is a test to verify the antiseptic effect of preservatives and it is a test to check the antiseptic effect and the level inhibiting growth of testing strains by observing increase and decrease of testing strains for 28 days after engrafting the testing strains in the testing sample for checking its antiseptic effect. The testing period requires about 2 months.

Percutaneous absorption
Percutaneous absorption is a process in which active ingredients in contact with the skin is absorbed through the horny layer or other routes, finally reaching the dermis and the blood vessels. To measure the skin absorption ratio of the medicine and the ingredients within the cosmetics, percutaneous eruption test, in which diffusion cell is employed, is used.
