Quality criteria for accurate shole-cell recordings:

• high seal resistance (Rseal>1GOhm)
  Rseal = 2.7GOhm(n=76, Kv1.5)
• high and stable membrane resistance (Rmem>500MOhm)
  94% stable whole-cell after gigaseal (Kv1.5), continuously monitored
• low access resistance (Ra<10MOhm
  Ra=9.92MOhm(n=47, HEK hERG cells)
• stable whole-cell configuration for at least 15 min

The CytoPatch^TM Pipette resembles a "real" patch pipette:

• glass surface for real gigaseals
• shape and diameter (ca. 2μm)
• no contamination of the tip of CytoPatch^TM pipette: positive pressure until sealing
• Rpip ca. 8MOhm (Ver.1) or ca. 4MOhm (Ver.2)

• high data quality due to stable gigaseals
• robust whole-cell recordings → high success rates and high through-put

These are contacted and filled via penetracting cannulaes, that are also used for cell and compound injection into the transport channel, pressure application for cell positioning and electrical contacting.
